Abstract
This study was aimed at adapting a DNA extraction protocol by Araucaria angustifolia leaves, and testing the informative capacity of RAPD markers for genetics diversity analysis in natural populations of this species. The extraction method was standardized by eight tested protocols and it was possible to Obtain good quality DNA for RAPD reactions. The OD260/0D280 ratio ranged from 1.7 to 2.0 in 80% of the samples, indicating that they had a low level of protein contamination. The RAPD markers demonstrated their capacity as a useful tool to genetic studies, since the estimate of a genetic diversity (Shannon index H=0.50) higher than that obtained by other authors with isozyme markers (H=0.29) and PCR-RFLP (H=0.0) for the same natural population of the Ecological Municipal Park, Lages, Santa Catarina, Brazil.Downloads
